HFWG participants demonstrated a significant (P < 0.005) increase, of at least 20%, in fecal acetate, total SCFAs, and indole concentrations, when contrasted with the control groups. Following WG treatment, a marked (P < 0.0001, 2-fold) elevation of the ileal interleukin 22 (IL-22) to interleukin 22 receptor alpha 2 (IL-22RA2) mRNA ratio was observed, which prevented the HFHC diet-induced increase in ileal protein levels of indoleamine 2,3-dioxygenase and phosphorylated signal transducer and activator of transcription 3 (pSTAT3). Human and livestock fertility can be significantly impacted by ovulation disorders. Female rodent ovulation depends on the luteinizing hormone (LH) surge, which is a consequence of kisspeptin neuron activity in the anteroventral periventricular nucleus (AVPV). ATP, a purinergic receptor ligand, potentially acts as a neurotransmitter, stimulating AVPV kisspeptin neurons to elicit an LH surge and consequent ovulation in rodents. Administration of the ATP receptor antagonist, PPADS, to ovariectomized rats treated with a proestrous dose of estrogen, when delivered into the AVPV, prevented the LH surge and led to a decrease in ovulation rates in those animals. The administration of AVPV ATP to OVX + high E2 rats caused a surge in LH levels during the morning hours. Critically, the application of AVPV ATP did not elicit an increase in circulating LH levels in Kiss1 knockout rats. Along with the previous points, ATP substantially enhanced intracellular calcium levels in immortalized kisspeptin neuronal cell lines, and concurrent administration of PPADS countered this ATP-stimulated calcium elevation. During the proestrous stage in Kiss1-tdTomato rats, a substantial increase in the number of AVPV kisspeptin neurons immunoreactive for the P2X2 receptor (an ATP receptor) was found, as visualized by tdTomato, linked directly to the estrogen level. Ovulation is proposed to be initiated by hindbrain ATP-purinergic signaling, which activates AVPV kisspeptin neurons, as these results suggest. Adenosine 5-triphosphate, acting as a brain neurotransmitter, was shown in this study to activate kisspeptin neurons within the anteroventral periventricular nucleus, the neural circuit generating gonadotropin-releasing hormone surges, utilizing purinergic receptors, leading to a gonadotropin-releasing hormone/luteinizing hormone surge and ovulation in rats. Furthermore, histological examinations suggest that adenosine 5-triphosphate is probably produced by purinergic neurons within the A1 and A2 regions of the hindbrain. Future therapeutic options for hypothalamic ovulation disorders in both humans and livestock may stem from these research findings.
